The Queen could appoint anyone PM, but the point is that the PM needs to form a government. If she were to choose a Conservative instead of Labor when Blair resigned he wouldn't have had a governing majority.
IIRC in 1940 after Chamberlain resigned there was some question whether the King would call Churchill or Halifax as PM, and it was basically his decision. But I think Churchill was more enthusiastic.
