Put the OS and apps on one volume, that you just leave alone. It will be mainly smaller files, so let OS X manage it.
Put your big work files, videos etc, on a different volume or (better) a totally different HD.
Exactly my view:
80GB internal - OS, apps & temporary + work files 3x120GB internal - software installers, data & audio books 250GB FW ext. - music 120GB FW ext. - films
I just found a 120GB disk I'd forgotten about which I can use for temp. storage/erase/backup cleaning.
