sure.

User directories in OS X can be moved to different hard drives; follow the directions in the OS Hacks section at <http://www.bombich.com> .

Just be careful; if your second drive has spaces in the name, make sure you put the destination path in quotes in the ditto command you'll use in Terminal when you do that.

Programs can be moved, with one caveat. Just make an alias to a folder on another drive in your Applications directory and put all non-OS installed programs there.

However, due to the way Apple updates your /Applications directory, the same tricks used for moving the /Users directory to another volume cannot be used. However, all the programs you install yourself could be put on another drive and symbolic links placed in the /Applications directory to make it look like they're all in that one directory. (Symbolic links you'll learn about if you move your Users directory. They're Unix' version of an alias. They look the same in the Finder (an icon with a little curved arrow) but operate differently on the OS level.)
