Select one of the media files in the finder, do File/Get Info, on the Open With menu in the dialog box select iTunes, and click on the Change All button. To open one specific file in Real Player (or whatever) control-click on the file and select the appropriate program from the Open With list -- but you have to go through the Get Info routine to change the default.
