Defragging is a vastly over-rated cure-all; it will not fix an ill drive, nor do drives become ill because of fragmentation. It is merely a performance thing.

If the drive will not mount, and Disk Utility repoirts an error on the drive, something is wrong with the drive or the drive contents. Time for bigger guns, like Disk Warrior.
