Hold the Shift key at startup to Safe Boot. This will rebuild all the System cache files. You don't need to actually login under Safe Boot, simply press "Restart" and boot normally, it may be fixed hopefully. If not, make sure the HD isn't too full. Something like Disk Inventory X can show you what's huge and what's small, and perhaps there's some huge file that's hidden, sometimes certain log files can grow huge if there's a problem.
