No. If you run out of runes of life, you get marks of death instead. You can't advance skills while you've got marks of death, if I remember rightly, and they cost twice the experience a rune of life does. Buying a rune of life is 16 experience. Removing a mark of death is 32. Always keep a rune or three around.
