Since I have to do something alike in the near future I have considered using time based filters to minimize the "vulnerability window". Although the solutions presented by others seem sound, backing up a Windows 2000 machine in such a way that you will be able to do a disaster recovery requires software like CA's ARCServe or Veritas BackupExec and therefore opening up all sorts of nasty ports to the PRO where the backup device resides Still a dedicated (locally attached) tape drive is by far the best solution.
