I have successfully changed IP addresses on the GNAT Box without rebooting. I'm not sure if GTA made a change to the GNAT Box after the manual was printed, or if someone just thought "better safe than sorry".
There are probably issues if you try to change the IP address of the interface that you are connected to by GBAdmin or the Web interface, though.
