Five to just before she's due to deliver.  You want to leave the
pups with her as close to six weeks as possible; yet not have
dad re-breed her.

Another thing, if she's tending pups and carrying, her second
litter will be a delayed thing; so I would go for at least five weeks
before removing dad and the boys.

Others with lots more litters under their belts will probably also
offer their expertise...
