no. There is no API exposed to the 'My Maps'

You can export features in KML format. Or indeed import a KML file -
but its a manual process.

But you can have your site update a database with your locations, and
then display the positions on a Google Maps API based map (and if you
create a KML file or GeoRSS feed then it can be displayed easily on
Google Maps too)
