Your question can mean a lot of things, depends on what you mean with "best route".
You can mean the travelling salesman problem that has been asked numerous times and got several answers. In this case try to search Google because the group's search seems still broken. http://www.google.com/search?q=site%3Agroups.google.com%2Fgroup%2FGoogle-Maps-API+travelling+salesman&ie=utf-8&oe=utf-8&aq=t&rls={moz:distributionID}:{moz:locale}:{moz:official} You can also mean if there is an alternative route available in the API as it is in Google Maps for a couple of times now. In this case the answer is no and you can post a feature request here http://code.google.com/p/gmaps-api-issues/issues/list You surely can mean something completely different with "best route" that is not easy guessable.
