This isn't currently possible using gregorio, but it's planned for the  
future.

As noted on http://home.gna.org/gregorio/gregoriotex/details , the  
format of the initial is governed by \greinitialformat (or  
\grebiginitialformat, in the case of an initial spanning 2 lines). If  
all you need is to change the size, some LaTeX like this should be ok:

\def\greinitialformat#1{%
{\fontsize{50}{50}\selectfont #1}% whatever font size you need
}

I think the main-lamed/lualatex files used with the Populus Sion  
example in the Gregorio download also show an example of this.
