What version of Windows are you using. On my XP machine Gregorio is installed to C:\Program Files\gregorio. TeXlive installs in the same location you indicated.

Have you checked the components individually yet? If not, try typesetting a simple document using lualatex (i.e. one using one of the basic document classes, no packages, and with just a short blurb of text as the content. If you can typeset that, the you can be fairly sure that TeXlive is installed properly and working.

Next, try running Gregorio from the command line on one of the examples taken from the website. This should tell you if Gregorio is working.

If both are working, then the likely culprit is in the TeXworks configuration (assuming you're using TeXworks to edit and typeset your files). If that's the case, let me know and I'll walk you through that process.
