Are you sure nothing happens?

In the case of GrowlTunes, look in your menu bar. When it's running,  
you'll see a pair of headphones. That's its status item, which lets  
you quit it (among other things). After you quit it, launching it will  
bring back the headphones.

Applications can only post Growl notifications when they're running,  
for the same reason why your lawn-mower can only cut grass when it's  
running. If GrowlTunes isn't running, you need to launch it. If it's  
running and you remove it from Growl's list, you need to quit  
GrowlTunes and re-launch it.
