If you set your notification preferences to play an alert sound for certain applications, they only work part of the time. Namely, those applications that do not customize the icon or title (a good working example is "Notify", the free Gmail notifier), or only use a single icon and title. For applications that use multiple icons or titles (like Hardware Growler or Tweetie), no notification sound is played.
To reproduce, try using Hardware Growler. Set a notification preference in the Growl control panel to play a sound for every Hardware Growler growl. Then, mount or eject a drive, or plug/unplug your Ethernet cable, or power cable. You will not hear a sound. Update: Upon further testing it seems that SOME events actually do play a sound. For example, my Hardware Growler will correctly play the alert sound when I connect or disconnect my Airport network connection. But for all other hardware events (USB, power, disk mount), no sound is heard.
