There have been exactly two changes in GrowlMail between b2 and b5.
One didn't change anything (__OBJC2__ is always defined when we target
Leopard, which we have for all of the 1.2 betas), and the other
doesn't affect you (since GrowlMail is not detecting a change in
Mail's behavior and disabling itself).
So, they did conflict back in b2. If you had the same version of
MailActOn installed at the same time and didn't have a problem then,
consider yourself lucky for that.
On the theory that GrowlMail is the broken one, here's a build
containing a possible fix:

If this doesn't fix it, I must assume that MailActOn is the broken one.
