It's not supposed to. It should be in the applications list in the Growl preference pane in System Preferences.
If you don't have the Growl preference pane, then you need to install that from the Growl 1.2 disk image. It doesn't come with GrowlMail. (Conversely, *don't* install GrowlMail from the Growl disk image—it's an older version that doesn't work anymore.)
