As far as I can tell, GrowlTunes works fine with iTunes 10. The appearance that
it doesn't—including my own previous belief that there was a problem—is
probably caused by a bug fix in 1.2.1:

If iTunes—any version—is the active application, GrowlTunes will not notify.
This is intentional.
I've just closed the other bug report. We'll reopen it if and when somebody can
show steps to reproduce the problem with iTunes in the background.
