It's harmless to have both of them, but you probably should remove the 
“unknown” one, just for cleanliness's sake (and it may also make startup a 
*tiny* bit faster).

The “unknown” entry is unknown because that GrowlHelperApp no longer exists. 
You probably got Growl installed by Dropbox or something, and whatever 
installed it put it in the wrong place. The Growl Installer package (when you 
upgrade) deletes it from there and installs into the correct place. The 
“unknown” entry is the old one, in the wrong place, that the upgrade deleted; 
the “application” entry is the newer one that is actually used.

So you should leave the “application” entry and remove the “unknown” entry.
