So applications will add themselves automatically if they work correctly.
The problem is that for Skype I believe you will see this problem:

Where Skype hasn't kept their Growl support up to date. Please contact
Skype, and feel free to try the workaround in the very well documented blog
post on ninjadesk.

Apps that are part of os x (mail, itunes) will need something else to work
with Growl. iTunes needs GrowlTunes, which you can find on the 1.2.2 disk
image for now. Mail will need GrowlMail, which you can find at
http://growl.info/growlmail/ . The developer on GrowlMail is also the Lead
Developer for Growl, and he's working on getting a better web page up for
GrowlMail.
