For iTunes you'd want GrowlTunes (or something else). iTunes does not support Growl natively, just like any other Apple application does not. So you need something to work as an intermediary.
GrowlTunes is available via the Growl 1.2.2 disk image. Spotify unfortunately implemented Growl support in a way that doesn't work with 1.3, I think they are assuming that Growl is a prefpane, and it's not for Growl 1.3.x. I'm going to try having Growl 1.2.2 installed but disabled, and Growl 1.3.1 running to see if that works, but that'd of course be an ugly workaround until Spotify gets things fixed.
