for Mail you'd need to either use AppleScript on specific mail rules,
or GrowlMail 1.3.2 http://code.google.com/p/growlmail/downloads/list

for iTunes you can grab the latest version of GrowlTunes from the
1.2.2 disk image, or use GrowlTunesPlugin 
http://github.com/rudyrichter/growltunesplugin
we are currently in the process of rewriting GrowlTunes.app to bring
it out of the dark ages and make it a more useful utility.
