It would need to remain open. Growl would not do the mail checking. Only Mail.app does that and Growl picks up on that. Also since I didn't see you mention it you would need to use the GrowlMail.bundle aka GrowlMail for Mail.app https://groups.google.com/forum/#!forum/growlmail-discuss is the discussion section or more info here http://growl.info/growlmail/
