There is no way to lock multiple task squares.

We usually suggest to people they not work on squares that have someone working next to them to avoid conflicts. So it can be partially addressed by training or task instructions.

There is a way to use custom task "squares" so you could make task "squares" that are a length of road that one person could check out.

I have not done this, but it is possible. There are some examples in the HOT Tasking Manager.

The other thing to keep in mind is that if you are using the main OSM live database, people could edit a road even not using the tasking manager. Not very likely depending on where you are editing, but it is possible.
