You've got a lot of things jumbled up in that question, Jake. :-) OK, to connect to CICS Transaction Server you need to connect, so yes, you will need physical network connectivity of some kind. The preferred way to do that is with an OSA Express adapter, yes. In the past there were many channel-attached networking options, but those options are generally fairly antiquated at this point. Presumably you already have physical network connectivity to your mainframe. Otherwise you would have a very "interesting" mainframe. :-)
You can use SNA protocols between Microsoft Host Integration Services and z/OS, assuming your network between those two will route SNA. You can configure Enterprise Extender if you wish as your SNA vehicle, and there are often good reasons to do that (such as network routing limitations), but it's an option. For the record, I've written previously -- and rather bluntly -- about the utility of Microsoft HIS (with or without BizTalk). You can find those past writings fairly easily.
