I'm a bit puzzled about this question.
The reason is that it not clear what you mean by "coupling" and how you
differentiate "machine learning" from "AI".

As far as I understand your processing goal, you need some kind of
particle detection to achieve the counting of specific shapes in images.
  From my understanding this is a classification task and there exist
plenty approaches, even classical ones.
