Probably because the 'main host' on the server has the web messaging contents. When you go to the hostname or IP-address of the 'main host' (IP-address of the server?) in your browser, you can login with i.e. "john@sales.mydomain.com"
 
Please note that you have to enable web access for the user "john" at the virtual domain.

Just make a new domain with the domainname "sales.mydomain.com", make a MX-record in your DNS entry for your domain pointing to your mailserver, i.e. email.mydomain.com.
