It depends on how quickly your drive gets fragmented.  We use diskkeeper and
it runs at night and it seems to keep up very well only running in off peak
times.  We process about 100K messages a day and also host mailboxes on this
server, but the mail boxes are forced to stay under 10mb so that helps.

My concern if you were defragging during peak times depending on your box IO
speed you could introduce a slowness that may be undesired.  What are you
using to defrag?
