Total of all messages for a single user. i.e. 10MB limit, once a box reaches that size, all further messages are either rejected by the smtp service with an appropriate 'over limit' message, probably a 500 series code, or alternatly by a plugin that deletes all further messages and sends non-delivery emails to the senders about the box being over limit.
