I do not know how to lock screen rotation using VoiceOver, but, here is how to 
do it using the usual user interface:

1. Double-tap the Home button, revealing the App Switcher
2. Slide the App Switcher from left to right, revealing the music controls
3. Tap the leftmost icon (the one with a circled arrow in it) to lock your 
orientation. Tap it again to unlock it.

If you are sight-impaired, I suspect that VoiceOver will announce these steps 
(but I do not know for sure). Does that work for you?
