Real digital signatures don't work with setAbsolutePosition(),
so I guess you're talking about a fake signature: some image
put as an image on a PDF. (Fake in the sense that it hasn't
any legal value, as opposed to a real digital signature that
is accepted as a valid signature by many countries.)

If you work with a real digital signature, then there's a digital 
signature field.
It's easy to get that real signature field.
If there's no signature field (nor any other field that marks the spot),
then you can't find out where that line is.

Use a field to mark the spot.
