To put my 2 cents in:

Sendmail on all unix platforms does this by checking a config file to see
which domains it should accept mail for.  You could do the same thing with
James.

To handle the mailboxes for multiple domains with James, in the userlist you
would add a domain field so that first you would check what domain you are
receiving then if the particular user existed in that domain.

MX records on the Internet visible DNS server for your domains would point
at your James server for instance:

Domain1.com
MX  james.domain.com

Domain2.com
MX james.domain.com

Domain3.com
MX james.domain.com

etc. , etc.
