You can definitely modify/create .xreg files and have the changes picked up by the registry service. Jetspeed's registry service is configurable in JetspeedResources.properties:

Since Jetspeed attempts to load Portlet classes when the registry service loads .xreg files, it stands to reason that you could add a jar containing a new portlet class by using your server's hot-deploy mechanism, and it would be picked up by Jetspeed in addition to the new .xreg entries.
