Sometimes your keyboard gets reset to another keyboard language. Try pressing Shift+Control until it say US, then try using them again. Also, if you are using a USB keyboard, try unplugging the keyboard and plug it in again. If it's a laptop, you may need to look in the manual because sometimes those keys can be changed to perform other things other than as function keys. Another thought, if you are in word and you press f12, andthe "save as" dialog box comes up, or f7 and the spell checker loads, then the function keys are working but the problem is with the control, alt, Windows, shift, or Insert key. Could your number lock be on?
