I've been having this issue and have played around with a few things to try and
work out what is happening, I still get this issue after resetting my spectacle
and clipboard settings to default (with the button in the bottom of the
settings for both)

Basically the way I get this issue to happen pretty consistently is I have
'capture region' bound in my system settings for spectacle, if I select a
region that would result in high file size (not just large file dimensions but
lots of detail in the image) then I hit ctrl-c to copy that image into my
clipboard and this is when it breaks. I can see the image in my clipboard
history but if I paste there is no image in my clipboard. If I select another
item in my clipboard history and then click back onto this new image, it
suddenly copies fine again and is able to be pasted.

Interestingly I don't get this issue if I manually run `spectacle -r` and
select the same large image that would break before when using the binding,
running it manually shows the image I copied in the spectacle window (unlike
the shortcut) and this appears to go into my clipboard just fine every single
time I've tried it.
