You need to configure this on the tablet driver settings level. How that is
done, depends on the OS and the tablet brand and model. Configure the pen
button to send ctrl-z in the tablet configuration utility that comes with your
OS. Vanilla Ubuntu uses Gnome these days, which comes with a suitable utility.
