If you're leaving the motherboard in the case with the switches, lights and speaker hooked up you don't need to hook up the drives or perhipherals. But in the interest of being safe and thorough, I would disconnect everything (drives, cdroms, etc) except the power switch and speaker and pull out all the cards. Then when you attempt to power up the motherboard, if it does come on you should hear a series of beeps (usually one long followed by three short) that indicate the board powered up but it is missing video plus the fans for the power supply and the CPU will come on.
