What OS are you on? libusb does not seem to work on 64-bit Vista machines.
Does the NXT device appear in Control Panel>System>Hardware>Device Manager? It
should appear under Lego Devices as something like "Lego Mindstorms NXT in
firmware update mode".

Typing >C:\lejosdl Tune.bin in command window gives "NXT not found. Is it
properly plugged in via USB?"
