In @rst and @md nodes, probably.  I haven't tested it myself.
3) can external files (pdfs, etc) be linked to nodes, so that by clicking on their link the external file is opened?
Yes, by using a @url node.
4) do Python plugins have access to the whole program API or just to a restricted subset of commands?
The whole API. It's quite extensive, too: http://leo-editor.readthedocs.org/en/latest/
5) can key shortcuts be defined for all commands in the API (or at least, for all menu commands)?
Probably, but that's a *lot* of keybinding.
6) can the user freely redefine key shortcuts? (without having to go to the program's source).
This is all handled in myLeoSettings.leo
