I get the same.  I suspect this is now a "feature" of windows' arcane 
permissions settings.  The simplest thing is to run lilypond from the command line, with 
an explicit path to the program - something like:

C:\"Program Files (x86)"\LilyPond2.22.0\usr\bin\liliypond

You can either run this from the location of the ly file (not the desktop!) or 
have an explicit fully stated path to the ly file. Alternatively you could add 
the path to the lilypond .exe to your path statement.
