I assume you mean in the authoring environment, not via Lingo.
Set the focus on the score (ctrl/cmd-4).
Now look at the gray horizontal bar that has the frame numbers on it. In
the area above, if you have one row of cells, click the "++" icon in the
upper right so you have 6 rows. (Actually, the "++" icon has the "+" signs
stacked vertically, but that's kinda hard to do in email--you get the idea).
At the very top of the score, just below the title bar, is a row that's not
divided into cells--that's where your markers go. Single-click in that row,
above the frame you want to mark. A down-triangle will appear, with the
label "New Marker." You can type your label name in there.
You can grab the label and move it forwards and backwards. To delete it,
simply drag it down, out of the label row.
