x3270 is an X application. c3270 is curses based, it runs in character mode. s3270 is for writing applications against a 3270 data stream.
So, basically: if you're running X, use x3270, and if not, c3270. x3270 is my favorite 3270 emulator, free or otherwise. I have been known to install Cygwin and XFree86 on Windows just so I could use x3270 there.
