You wouldn't need to use nat with squid unless you're trying out 
transparent proxying or are trying to allow internet access to your 
internal network that is on a private ip.

If you're trying out transparent proxying with squid, you need to download 
the latest 2.4 stable squid series and compile it with support for 
iptables. Also, if you're using nat, iptables is a better choice than 
ipchains, simply because it's more capable. There's a howto on transparent 
proxying which you can find on google which will detail the steps required 
in detail.

Transparent proxying has it's benefits- you don't need to configure all 
your clients to use a proxy.. it happens automagically, and it's 
disadvantages- dns queries aren't cached by a transparent proxy.

Ipchains and iptables are used to improve the security of your box.... 
using it wouldn't really improve any speeds.
