I hope you are substituting in text files...

sed -e 's,/usr/bin,/usr/local/bin,g'

Or, better still,

sed -e 's,\(/usr\)\(/bin\),\1/local\2,g'

ie, If you use another separator for sed, you don't have to escape
the slashes...
