Actually, you do not need a DOS partition to run windows program under wine.

The type of partition is irrelevent. You even can create a new directory on your
existing file system and use that if you wish.

The structure of the directory may or may not be important. Some windows
software are known to look for the "windows\" directory and may complain if it
can't find it.

Incidently, if you have never tried wine before, you might want to give it a
test run first before you put too much time into it. Wine is still in
developement stage and is incredibly buggy and unreliable.
