Yes and No.  
IIRC Windows starts the Graphics Desktop Manager, then the first instance of Windows 
Explorer as the desktop widgets - I expect this is what you're getting at.  Subsequent 
instances will run as file managers.  You can stop all instances of Explorer and run 
another desktop widget set - or no widgets at all, as long as you have some means of 
starting programs (say cmd.exe).  Under it all, GDM supports the Desktop, a bit like X.

Yes, Explorer will run with wine.  
No, It won't become a desktop and support other applications; it will be a file 
manager, just like on the Windows desktop, but without the lovely cut'n'paste and 
drag'n'drop support from the desktop environment.  I really miss those . . .
