Do you known the free (GPL) software named sympa ? It allow very large
lists and a lot of configuration. One of them allows to create distribution
list using an external database : sympa can contact you compagny database
server and dynamically extract email of a category of employee.

Sympa include a web interface for list administration and list subscribers
including list archive with access control etc etc.
