I know how to set up an IBM Client Access connection via ODBC, is that what you need?
If so: 
1. First make sure you install Client Access on your server.
2. Start/Programs/Administrative Tools/ODBC Data Source Administrator
3. Click on the Tab System DSN.
4. Click on the Button "Add".
5. Select "Client Access ODBC Driver (32-bit)" (Should be the first on the list, find it if not), Then click "Finish".
6. The Client Access Express ODBC Setup (32-bit) box will pop up.
7. On the General Tab fill out the following:  Data source name (Whatever you want), and AS/400 System (Select AS400).
8. On the Server Tab fill out only the Default libraries (This has to exist on the AS400 or else nothing will work).  A library on the AS400 is like an Access DB, and a library has many files like Access DB has many tables.
9. Click "OK".
10. Go into the ColdFusion Administrator under ODBC Data Sources.
11. Click on the Data Source Name you just created which is step # 7 above.
12. Enter the Username and Password for the ColdFusion Login.  This is important.  If your AS400 requires this Username and Password, most likely it will, and you forget to fill this out...you're going to have hang time problem...like forever, until you fill out the dialogue box that prompts you for the username and password which is done by the server sneakily. 
13. Click the Update button.
14. Go back to your ODBC Datasources and "verify" the Data Source Name you have just created. 
 
If you get "The connection to the data source was verified successfully." your done!  If not, check to see if the AS400 is connected to your network.

We use IBM Client access as an ODBC datasource where I am.  We have a part-time AS400 guy come in every once in a while.  I can ask him questions if you have any particular ones.
