For "Download content from distribution point and run locally"....
Set-CMApplication $ApplicationName -SendToProtectedDistributionPoint $True

and "Allow clients to use a fallback source location for content"...
Set-CMDeploymentType -AllowClientsToUseFallbackSourceLocationForContent $true 
-ApplicationName $ApplicationName -DeploymentTypeName $DeploymentTypeName 
-MsiOrScriptInstaller

Obviously these are examples and will need to be modified to work with your 
script.

So the script below now working fine for ticking the “Allow clients to use a 
fallback source location for content” box.

But I also need it to set the dropdown box to “Download content from 
distribution point and run locally”..  and on the Distribution Settings page, 
“Distribute the content for this package to preferred distribution points”.

I can’t find any powershell cmdlets for these,  any ideas?

I need to set Fallback on all my applications, and a quick google found me the 
below page, however the script doesn't work at all for me, and I’m yet to tick 
off the “learn powershell” from my to-do list..
